Mitotic spindle is necessary for mitosis of the cell. If it is damaged, then chromosomes will not be able to attach to it due to which they cannot be aligned toward the center during metaphase. And they will not be able to continue the next stage of mitosis, which needs chromosomes to reach opposite ends of the cell. Hence drugs like Vincristine damages the mitotic spindle effectively blocking the cell division.
DNA template is necessary for the formation of mRNA, which takes the information from DNA and takes that to ribosomes to instruct them for building proteins. Drugs like Doxorubicin blocks the DNA and stops the synthesis of mRNA due to which essential proteins could not be generated and cells die. These drugs are harmful to both cancer and human cells.
