What is the significance of Uncle Ben's death for Spider-Man?
Peter has acquired great gifts and become Spider-Man. He is meant to meet his Uncle, but gets carried away with his powers and has entered an underground fighting competition; Ben is murdered while he waits for Peter. This event creates a rage in Peter that turns to him allowing the death of Ben's killer rather than attempting to save him and have him tried for murder. The significance is that Peter is still very much human but doesn't live by the same rules as the rest of us. His power is his responsibility, and we see his struggle to use it completely for good.

How does the Daily Bugle gets photos of Spider-Man?
J Jonah Jameson is the editor in chief of the Daily Bugle and he wants photos of Spider-Man, but no one can get anything more than a blurry shot of him in the distance. Jameson wants to sell papers so Peter decides to get in with Jameson by telling him he can get photos of the web-slinger exclusively for the Daily Bugle. Peter then sets up his camera and changes into his Spider-Man suit and sells his photos to Jameson exclusively, who then wants to pay him very little for his shots.

How does Mary Jane become a target for the Green Goblin?
Norman Osborn has become the Green Goblin and fights Spider-Man. Soon after he finds out the Peter Parker is Spider-Man when he sees a cut on him from the fight they had while both were in their suits. Mary Jane becomes the Green Goblin's target once Norman discovers that Peter is in love with her from his son Harry. Harry has told his father about this because he was dating Mary Jane and caught her holding hands with Peter, which absolutely crushed him. So he came to his father consolation, and his dad used this information to get to Peter and ultimately Spider-Man.
