Sleep is a symbol of innocence because the innocent quickly find the state of slumber without things that cloud their mind.... innocence sleeps without guilt. In Macbeth, Macbeth's act of cold-blooded murder leaves him guilt ridden, he has lost his innocence and can no longer find sleep.
