Why does poole believe that the person in the laboratory is not Jekyll?

Poole admits that he believes there has been "foul play", but refuses to go into specifics. Poole says that a man has been pacing back and forth, and asking for certain drugs on a piece of paper, never being fully satisfied and sending them back. He thinks the man might be a murderer.
